Description

The USDA, Agricultural Research Service in Albany, California has a requirement for bi-directional sequencing of polymerase chain reaction (PCR) amplicons [PCR reactions will be performed in-house and no assembly or analysis will be required; these amplicons will be shipped to the vendor for processing]. The sequencing process must be performed in a fully-automated production sequencing facility that tracks the quality and genealogy of all samples in real-time, on a well-by-well basis using a fully-integrated laboratory information management system (LIMS) to ensure accurate genealogy. The quality of pass rates must be 85% - 90% with average PHRED 20 read lengths of 650 ? 700 bp. Forward and reverse sequencing reads must be performed within the same sequencing plate to assure a high linkage rate. Detection of sequencing products must be performed on an ABI 3730 so as to share data with on-going sequencing project. Chromatograms, PHRED quality scores and FASTA sequence files must be delivered via FTP. Turnaround time must be within 10 business days for 10,000 or less amplicons.
