Description
The National Cancer Institute (NCI), Laboratory of Biosystems and Cancer (LBC), Center for Cancer Research (CCR), HIV and AIDS Malignancy Branch (HAMB) plans to procure, on a sole source basis, for Computational Studies for the Elucidation of a Structural Model for the binding of CCR5 Inhibitors with Debananda Das, Ph.D., 12903 Woodcutter Circle, Germantown, MD 20876. The acquisition is being conducted under the simplified acquisition procedures authorized in FAR Part 13. The North American Industry Classification System Code is 541990 and the business size standard is $6M. Background: The HIV and AIDS Malignancy Branch (HAMB) conducts laboratory and clinical research in AIDS-related malignancies, HIV disease, and viral-induced tumors. The principal aims of this translational research program are to develop novel therapies for these diseases based on an understanding of their pathogenesis. The Contractor shall utilize state of the art methodologies in answering question posed by biologists and help them interpret experimental data for protein inhibitor complexes. In addition, the Contractor shall work in the laboratory to successfully advance the research projects. The projects involved are: 1) Refining structural model of CCR5, 2) Inhibitor design to find lead compounds as inhibitors of CCR5, 3) Lead optimization of inhibitors to improve potency, 4) Structural models of HIV-1 components, in particular Gag protein, and 5) Peptide and small molecule design for protease dimerization inhibition. The computational methodologies used shall include homology modeling, protein structure refinement, molecular docking and scoring, molecular mechanics and dynamics, pharmacophore model elucidation, and molecular database searching. Period of Performance: Base year plus two (2) one-year options. The Contractor is the only person known to the NCI to perform this work due to his extensive experience and unique knowledge on computational methodologies that are essential for gaining valuable insights in the projects listed above. The Contractor understands the biology of HIV, the pathogenesis of AIDS, and the role of molecular modeling in analyzing experimental data and has the communication skills necessary to work effectively in a multi-disciplinary environment. The Contractor has unique qualifications and can immediately support our ongoing research projects by using computational docking and molecular mechanical methods for evaluating ligand protein interaction energies. This notice of intent is not a request for competitive quotation. However, if any interested party believes it can meet the above requirements, it may submit a statement of capabilities. The capability statement and any other furnished information must be in writing and must contain material in sufficient detail to allow NCI to determine if the party can fully meet the requirements herein. Capability statements must be received in the contracting office by 1:00 PM EsT on February 3, 2006.
