- The National Institutes of Health intends to negotiate on a sole source basis with Dr. Sugata Ray, Columbia, MD 21044 for the following services: 1. The contractor shall provide molecular biology services for functional genomics work relating to the role of platelet-derived growth factor D in the eye. 2. The contractor shall complete ongoing NEI work leading to the conditional knockout of the PDGFD gene in mice to elucidate the role of this factor in specific eye tissues, mapping of gene expression by IF, ISH and transgene expression. 3. The contractor shall use yeast 2-hybrid methods to identify binding partners and possible receptors for PDGFD in the lens and retina. 4. The contractor shall maintain mouse colony inventory by keeping breeding populations and culling old and unneeded mice, genotype for PDGFD-GFP transgenics. 5. The contractor shall compare native PDGFD and transgene expression during development for PDGFD-GFP transgenics. 6. The contractor shall maintain inventory, genotype for PDGFD knockout. 7. The contractor shall maintain mouse colony inventory by keeping breeding population and culling old and unneeded mice, genotype for H-retbindin transgenic mice for PDGFD knockout. 8. The contractor shall maintain mouse colony inventory by keeping breeding populations, culling old and unneeded mice, genotype for H-retbindin transgenic mice. 9. The contractor shall organize mice for microarray experiment to survey effects of transgene expressions in mouse eye for H-retbindin transgenic mice. 10. The contractor shall use relevant techniques including: PCR, DNA, Northern and Western blotting, yeast 2-hybrid methods and functional assays. 11. The contractor shall provide deliverables of genotyped modified mice and analyses of phenotype and developmental profiles to the NIH Project Officer. 12 The contractor shall carry out further molecular biology experiments implied by the results of expression studies, relevant to understanding disease and important biological processes. 13. The work will be performed in the Section on Molecular Structure and Functional Genomics, National Eye Institute, Bethesda, MD. 14. The period of performance shall be April 24, 2006 to April 23, 2007. This proposed action is for services for which the Government intends to solicit and negotiate with only one source. Interested parties may identify their interest and capability to respond to the requirement or submit proposals. This notice of intent is not a request for competitive quotations. However, all quotes received within ten days after the date of publication of this synopsis will be considered by the Government . A determination by the Government not to compete this proposed simplified acquisition requirement based upon responses to this notice is solely within the dicretion of the Government. Information received will normally be considered solely for the purpose of determining whether to conduct a competitive acquisition.
