Description
USAMRIID Statement of Work USAMRIID will prepare total RNA samples and send them to awardee for processing on Affymetrix GeneChips. Once the Samples arrive to awardee, the following protocols will be performed on each sample: 1. Sample will be analyzed for quantity and quality on the Agilent 2100 Bioanalyzer. If the sample meets the quality specifications (no degradation, no genomic DNA contamination, good 260/280 ratio, at least 1 ug total), the sample will continue to step 2. 2. Sample (1-5 ug) will be converted to cDNA using the standard reagents and protocols provided by Affymetrix. 3. The cDNA generated in step 2 will then be converted to CRNA, again using the standard reagents and protocols provided by Affymetrix. 4. The cRNA will be quantitated by spectrophotometry, fragmented and hybridized to the GeneChips overnight, again using the Affymetrix standard protocols. 5. Following the overnight hybridization, the chips will be stained and washed in an automated fluidics station, using the standard reagents and protocols provided by Affymetrix. 6. Chips will then be scanned and all data collected and stored in the CLF database. Data files will then be sent to USAMRIID for necessary data analysis. The data will be sent to USAMRIID either on CD or electronically. 7. All RNA, CDNA and cRNA regents will be stored at -80 degree C at the CLF unless the researcher requests for them to be returned.
