Description The National Cancer Institute (NCI), Division of Cancer Epidemiology and Genetics (DCEG), The Nutritional Epidemiology Branch (NEB), plans to procure on a sole source basis with the University of Bolton, Dean Road, Bolton BL3 5AB, UK for a high-resolution NMR analysis of saliva and esophageal cells. This acquisition will be processed in accordance with simplified acquisition procedures as stated in FAR Part 13.106-1(b)(1). Only one award will be made as a result of this solicitation. This will be awarded as a firm fixed price type contract. Period of Performance: Performance shall be for twelve months. NEB conducts intramural research in the areas of nutrition and cancer, genetics and cancer, and cancer prevention. The NEB is currently examining the hypothesis that differences in saliva chemistry are associated with the different risks of esophageal squamous cell carcinoma and other cancers. The NEB plans to measure saliva and esophageal cell chemical analytes using high field proton high-resolution NMR analysis. The purpose of this procurement is to obtain data on saliva and esophageal chemical composition and to test the effect of different methods of saliva collection, preservation, and analysis. The Contractor requirements are as follows: 1.The contractor shall complete high-resolution salivary 1H NMR analysis on 160 saliva samples; 2.The contractor shall determine the reliability and reproducibility of multi-component 1H NMR metabolic data acquired on the saliva specimens; 3.The contractor shall examine the utility of high-resolution 1H NMR analysis for frozen esophageal cells using 40 samples, to include cells analyzed by 1H magic angle spinning solid-state NMR and post-neutralized perchloric acid extracts analyzed by 1H NMR; 4.The contractor shall conduct post-analytic data analysis including principal components analysis to detect outliers or unknown clusters and multilevel component analysis to assess variance due to differences in sampling or oral health indicators. The current acquisition is based on the need to generate data on the chemical composition of saliva and esophageal cells using high-resolution 1H-1H TOCSY and COSY NMR spectra for saliva and the use of magic angle spinning solid state NMR on intact esophageal cells to aid in our investigations of the link between oral health and upper gastrointestinal cancer etiology. These methods offer numerous advantages over competing methods including flexible input samples collection methods and minimal sample processing. The NMR analytic method requires essentially no prior knowledge of the chemical composition of samples, yet can provide detailed information on both endogenous and exogenous chemicals in different biological sample types. The Centre for Materials Research and Innovation at the University of Bolton has previously demonstrated expertise in the analysis of human saliva samples using 1H NMR-linked chemometric techniques in the diagnosis, prognosis, and severity monitoring of oral diseases, and have thus demonstrated the ability to complete the desired analyses. The Centre's expertise extends to the assessment of sample integrity and intra-individual variation in salivary chemistry. Furthermore, the Centre for Materials Research and Innovation has already developed the post-laboratory data processing and statistical methods necessary for full utilization of the generated data. The NCI knows of no other providers that can provide the desired services, both laboratory and data analytic, and that have already demonstrated valid measurements of human saliva. This is not a solicitation for competitive quotations. However, if any interested party believes they can meet the above requirement, they may submit a statement of capabilities. All information furnished must be in writing and must contain sufficient detail to allow the NCI to determine if it can meet the above unique specifications described herein. An original and one copy of the capability statement must be received in the NCI Office of Acquisitions on or before 11:00 AM EST on February 18, 2010. No electronic capability statements will be accepted (i.e. email or fax). An original and one copy must be sent to the NCI Office of Acquisitions at the address stated above. A determination by the Government not to compete this proposed contract based upon responses to this notice is solely within the discretion of the Government.
