Description
The U.S. Department of Commerce (DOC), National Oceanic and Atmospheric Administration (NOAA), National Marine Fisheries, Protected Resources Division, located in St. Petersburg, FL intends to acquire services for investigations into the population genetics of largetooth sawfish, Pristis perotteti. DESC: This is a notice to acquire services for investigations into the population genetics of largetooth sawfish, Pristis perotteti Purpose: To investigate whether historically largetooth sawfish individuals were seasonal migrants or made up a US genetic breeding unit of the population of largetooth sawfish. The government will also make a range-wise assessment of population genetic diversity in the Western Atlantic basin, to examine the potential for a population bottleneck. Scope Proposed work: Tissue samples from U.S. collected specimens of Pristis perotetti will be obtained from dried rostra archived in museum collections (N=41 have been confirmed in museums in the U.S.). Small (1 x 1 cm) pieces of each rostrum will be collected with a sterile razor blade. The sample will be thoroughly washed in water and alcohol prior to DNA extraction and only the internal part of the sample will be used. Samples of P. perotetti from Central America will be obtained from rostra held in museums and private collections. Samples from South America will be collected from wild captured specimens (V. Faria pers comm). Genomic DNA will be isolated from all specimens using commercial kits and/or protocols described by Phillips et al. (2009) for obtaining genomic DNA from sawfish rostra. Two mitochondrial DNA loci (ND2, control region [CR]), that preliminary work has indicated are variable in P. perotetti, will be amplified using PCR in all specimens. In some cases we will attempt to amplify the whole locus in a single reaction, but in situations where genomic DNA is degraded due to age we will sequence these loci in 250-300 base pair segments to construct the whole locus (~1000 base pairs each) for every individual. Sequences for ND2 and CR will be aligned and genetic diversity indices for the species will be calculated in DnaSP 4.0 (Rozas et al. 2003). A statistical parsimony network will also be constructed for the observed haplotypes in TCS 4.1 (Clement et al. 2000). Genetic differentiation (ΦST; Jukes-Cantor distances) between the sampling sites and test of their significance will be calculated in Arlequin 2.001 (Schneider et al. 2000), using AMOVA. We will characterize haplotype and nucleotide diversity for each sampling region and determine whether U.S. specimens are genetically depauperate relative to the others. We will test the hypothesis that U.S. sampled specimens constitute either a set of distinct haplotypes from those observed in the Caribbean, South America and West Africa or that there are significant differences in haplotype frequencies between the U.S. and all other sampled regions. The minimum requirements: The contractor must possess a Ph.D in biological sciences or related fields and have conducted genetics research on elasmobranchs. The contractor must have demonstrated a record of publication of results in peer-reviewed journals particularly as it relates to the conservation genetics of sawfishes. To be considered, firms must furnish detailed information concerning their education, experience and capability to provide the services and satisfy or exceed the Government's needs. Such information shall include, at a minimum, name, telephone number and address of concerns for whom you have furnished this type of services previously, and detailed narrative that described your facility and ability to provide this service. Interested parties must respond within (14) days after publication of this notice. The government knows of only one source Dr. Kevin A. Feldheim, Field Museum of Natural History that can provide this services. Dr. Feldheim has a decade of experience developing genetic markers for elasmobranchs and has published a large number of scientific articles in this research area. He has worked on the genetics of another sawfish (Pristis pectinata) developing microsatellite markers to study the population genectics of this species. He has also developed species-specific mitochondrial markers to aid in the conservation of this endangered elasmobranch. The timing is crucial to the government in making a final listing decision on the largetooth sawfish if the species is proposed for listing. Thus, timely completion of this project is imperative. The government invites firms that believe they have the capability to meet the requirement to response, providing specific information that will enable the government to determine whether or not to compete the requirement.
