Description National Institute of Diabetes and Digestive Kidney Diseases (NIDDK) plans to procure on a sole source basis with Molecular Devices, 131 Orleans Drive, Sunnyvale, CA 94089 for Spectra Max M5e with current SoftMax Pro software, user manual, dust cover and 1 yr. warranty and SoftMax Pro v6 The upgraded M5e provides additional detection modes of Time-resolved Fluorescence, Fluorescence Polarization and Glow Luminescence. In addition the M5e has HTRF (Homogeneous Time Resolved Fl1uorescence) assay technology. This technology has been applied to many antibody based assays including GPCR signaling (cAMP and IP-1), kinases, protein-protein interactions and is frequently used fordrug target studies. We anticipate that converting our current ELISA-based assays to this technology will increase sensitivity significantly and broaden our range of detection. Converting radioligand binding with the Tag-lite system can be used to assay our GPCR ligand binding as well. We have established a number of GPCR signaling FRET probes that we currently use solely on our microscopes; microscopic acquisition and analysis is required for initial studies but can be converted to plate reader format for large experiments. Our current plate reader is not sensitive enough to detect changes in FRET signal. The upgraded M5e has the sensitivity and would thereby allow us to perform multiple FRET-based experiments with much less hands-ontime and post acquisition analysis. The Fluorescence Polarization capability can be used to determine, for example, ligand binding to mouse brain membrane preparations. This technology is a nonradioactive GPCR ligand-binding assay and has been used successfully to detect low density receptor membrane preparations. Fluorescence polarization can be used for receptor agonist and antagonist screens as well. It will also allow for measure multiple samples of RNA simultaneously. This is not a solicitation for competitive quotations. However, if any interested party believed they can meet the above requirement, they may submit a statement of capabilities. All information furnished must be in writing and must contain sufficient detail to allow the NIDDK to determine if it can meet the above unique specifications described herein. An original and one copy of the capability statement must be received in the DK Acquisitions Branch on or before 11:00 AM EST on September 21, 2012. No electronic capability statements will be accepted (i.e. email or fax); an original and one copy must be sent to the DK Acquisitions Branch to the address stated above. A determination by the Government not to compete this proposed contract based upon responses to this notice is solely within the discretion of the Government.
