Description
MEC for MiSeq Sequencer System (Brand Name or Equal) Must integrate amplification, sequencing and data analysis in a single instrument with a footprint of less than 2 feet square. Must NOT require use of a dedicated ancillary amplification system or computing infrastructure. Must NOT require emulsion PCR. Must provide less than 8 hour turnaround time from purified DNA to direct reporting of variants. Must require as little as 50 minutes total hands on time from sample preparation to analyzed data. Must be paired-end capable and can support up to 2 x 150 bp read length. Must produce greater than 1 Gb of high-quality output data. Must provide accurate variant detection enabled by proven reversible-terminator sequencing by synthesis (SBS) chemistry. Must produce greater than 85% of bases with Q scores greater than 30 (1 x 35 bp). Must allow for competitive addition from a pool of all four reversible terminator nucleotides labeled with four different fluorescent dyes to eliminate homopolymer errors. Must support TruSeq custom targeted enrichment kits and assays. Must be able to interrogate 10s to 100s of targets per sample and read variable length amplicon sequencing spanning 100s to 1000s of base pairs to support field diagnostic sample identification and microarray assays. Must contain light-emiting diodes at 530nm and 660nm. Must be able to utilized small sample input such as: <1ug DNA for genomic DNA sequencing applications and <1ug total RNA for small RNA sequencing and mRNA-Seq applications. Must be able to multiplex samples in 96-well and 384-well formats. Must support application flexibility allowing for multiplexed sequencing of viral and bacterial genomes, as well as single-end runs for CHiP-Seq and small RNA applications. Must be able to be utilized for whole-genome sequencing, targeted resequencing, de novo sequencing, metagenomics, mRNA sequencing, tag-based gene expression, small RNA sequencing, ribosome profiling, DNAse 1 hypersensitivity site mapping, and paired-end mRNA sequencing, etc.
