- Portland VA Medical Center is seeking a contractor to provide GC/MS fatty acid analysis. Services will include the gas chromatography/ mass spectroscopy analyses of ~800 red blood cell (RBC) blood samples in order to meet specific aim #3 of Dr. Shannon's VACO-funded Genetic Susceptibility, Environment and Prostate Cancer Risk study. Determine fatty acid concentrations in ~800 RBC blood samples via the use of gas chromatography/ mass spectroscopy. The Government requires the delivery of results and interpretation of said results via email. Specifically, the following was proposed for the fatty acid analyses process: one hundred ul of red cells shall be mixed with 250ul of a Reagent1(methanol and KOH) in a GC vial and vortexed. Then 250ul of a Reagent2(Hexane) is added to the vial and similarly vortexed. Lastly, 250ul of a Reagent3(dyed and diluted 12N HCl) is added before transferring 50ul of the organic layer to an insert for GC analysis. The FAME extract shall be injected in a split mode (1:30) and will be separated on a Hewlett-Packard, Model 6890 gas chromatograph (GC) (Agilent Technologies, Wilmington, DE). The GC system is equipped with a flame ionization detector (FID), electronic pressure control (EPC), ChemStation software (Agilent), and automatic sampler (HP7673). FAME shall be separated on a 25m by 0.20mm i.d. capillary column (Agilent J&W19091B-102 Ultra 2) with 0.33um coating. The carrier gas is hydrogen at 60 PSI and make-up gas is nitrogen at 60 PSI at the tank. The injector and the detector port temperatures are set at 250oC and 300oC, respectively. The initial oven temperature is 190oC and then the oven temperature will be increased to 285oC at 10o/min for an approximate 9 minute run. Finally, the oven temperature will be raised at 60o/minutes to 310o and held for 1 minute. Contractor shall NOT have access to patient records or identifiers. No background investigation is required. All blood specimens shall be labeled with the study identification (code) number. All blood specimens shall be sent via overnight tracked delivery by the investigator's staff. No employee of the contractor will require access to Dr. Shannon's -80' freezer (in which our specimens are stored) or to Portland VA secure network drive (where we retain the link to study identifiers). This solicitation is set-aside 100% for small business. Bids will be due back on or about 12/28/2012.
