Description
STATEMENT OF WORK The purpose and scope of this research contract is to purchase scientific research equipment (Brand Name or Equal). Mouse Cardiovascular Imaging System (VisualSonics): The mouse cardiovascular imaging system is comprised of specialized echocardiogram transducer and analyses suite used to image the heart in mice. This is a powerful technique to noninvasively image systolic and diastolic cardiac function in mice with minimal pain and discomfort to the animals. The parameters that can be assessed using this system include: i) evaluation of LV and RV ejection fraction and stroke volume, ii) wall thickness, iii) pulmonary arterial pressures, and iv) LV and RV diastolic function (using pulse wave Doppler, tissue Doppler, strain rate imaging and speckle tracking). Mice (both wild type and genetically manipulated) are commonly used in several ongoing projects at the Providence VAMC and serve as an indispensible tool to evaluate the mechanisms underlying several diseases such as emphysema/COPD, heart failure, pulmonary hypertension and acute lung injury/ARDS, significant problems in the veteran population ad actively investigated at the Vascular Research Laboratory at the Providence VAMC. In addition, mice are used as preclinical models to test for efficacy of novel therapeutic agents in these diseases. In order to perform sophisticated analysis of systolic and diastolic right and left heart function generally invasive techniques using pressure-volume catheters are used. This technique has several limitations. The technique is associated with pain and distress requiring deeper anesthesia that can cause complications and loss of animals. Also, due to invasive nature, we are unable to perform these to assess the temporal progression of the cardiac dysfunction. Many of these limitations can be overcome by performing cardiac function analysis using the requested equipment. The mice are anesthetized using inhalational anesthetic that can be titrated for comfort, have quick recovery and we can perform repeated analysis during the time course of disease progression. Furthermore, we have the technical expertise to perform these studies. Dr. Choudhary, a VA funded PI, is an expert in echocardiography and has successfully performed these analyses. The VisualSonics equipment will be used in conjunction with the VisualSonics Vevo 2100 and imaging bench that is available at the Providence VAMC. Hence, the availability of the mouse echocardiography system would enable the investigators to acquire important data pertaining to cardiac function non-invasively with minimal pain and discomfort from a wide-variety of preclinical models of cardio-pulmonary diseases evaluating the underlying mechanism and therapeutic efficacy of compounds. The scientific equipment will be located in Research Building 9, Room 107. 1.The contractor shall work with the research team to carry out the services/requirements described below. 2.Requirements: The contractor shall provide the following scientific equipment request: VisualSonics Mouse Cardiovascular Imaging System to include: 1.Part # VS-11960: MS550D 55 MHz MicroScan transducer "Broadband Frequency: 22 MHz - 55MHz "Applications: mouse abdominal, reproductive, mouse/rat embryology, tumor imaging (up to 14 mm in diameter), mouse vascular, small rat vascular, some abdominal kidney) "Deep Focal Zone "Max Frame Rate: 557fps "Image Width: 14.1 mm, Image Depth: 15 mm "Image Axial Resolution: 40 um 2.Part # 11954: ECG-Triggered - Respiration Gated Analysis "Provides physiologically timed LV-analysis, supporting: "Single frame gating at systole and/or diastole in all 2D Modes "Cineloop acquisition for a user-defined # of cardiac cycles 3.Part # 12127: Anatomical M-Mode "Allows user to steer the sample volume to any angle rather than having sample volume in a strict vertical position "Requires M-Mode 4.Part # 11951: Color Doppler Mode "2D and color acquisition for blook flow velocity analysis using advanced digital Signal Processing techniques. "Requires PW Dooppler Mode 5.Part # 11846: VevoStrain Analysis Suite "Provides tools for myocardial strain, strain rate, and time-to-peak analysis "This feature also includes dynamic assessment of myocardial velocity and displacement. 3.Delivery Items in support of delineated requirements will be delivered over the time period from the effective date of contract, through the date of the last line item. 4.The contractor shall submit invoices in a timely manner to the VA upon delivery of requirements. 5.Performance under this contract is subject to all VA and VHA regulations and policy without limitation as to those now in effect or as may be established.
