- (i) "This is a combined synopsis/solicitation for commercial items prepared in accordance with the format in Subpart 12.6, as supplemented with additional information included in this notice. This announcement constitutes the only solicitation; proposals are being requested and a written solicitation will not be issued." (ii) The solicitation number is HHS-NIH-NHLBI-CSB-(HG)-2015-147-DLM and the solicitation is being posted as a request for proposal (RFP). (iii) The solicitation document and incorporated provisions and clauses are those in effect through Federal Acquisition Circular 2005-82, June 8, 2015. (iv) This is not a set aside, the associated NAICS code is 541711, Research and Development in Biotechnology and small business size standard is 500. The acquisition is being conducted in accordance with the procedures of FAR Part 12 and FAR Part 13. (v) Procurement Line item: 1. Create one (1) knockin mouse model using embryonic stem (ES) cell based conventional homologous recombination approach for U2af1 missense mutation (Q157R) conditional knock-in (loxP-mini cDNA-STOP-loxP cassette upstream of the point mutation), from design to Germline Transmission F1 mice, strain background C57BL/6. Quantity: One (1). Procurement Milestones/Time: Milestones: Part 1: Design/Creation of Targeting Vector, DNA sequencing verification; Timeline: 1-2 Months; Milestones: Part 2: ES cell electroporation and culture; Timeline: 2-3 Weeks; Milestones: Part 3: Positive clone isolation and screening; Timeline: 3-5 Weeks; Milestones: Part 4: Positive ES cell clone expansion and Microinjection; Timeline: 2-3 Months; Milestones: Part 5: Housing/Breeding of chimeras to F1 heterozygotes and confirmation of germline transmission via genotyping; Timeline: 2-3 Months; Shipping and Handling; Timeline: 2-4 weeks. Background: The National Institute of Health (NIH) is the nation's leading medical research agency and the primary Federal agency mission is to seek fundamental knowledge about the nature and behavior of living systems and the application of that knowledge to enhance health, lengthen life, and reduce illness and disability, conducting, supporting and making medical discoveries that improve people's health and save lives. The National Human Genome Research Institute (NHGRI) mission has expanded to encompass a broad range of studies aimed at understanding the structure and function of the human genome and its role in health and disease. The NHGRI Cancer Genetics and Comparative Genomics Branch seek to identify and study genes that contribute to cancer susceptibility and progression. They focus on inherited and somatic (body cell) mutations that contribute to cancer susceptibility syndromes and to the development of various aggressive cancers, including prostate, endometrial, bladder, lung, head and neck, and gastric cancers. They also study the relationship between genetic variation and cancer progression and pursue the discovery of both inherited and somatic variants that lead, specifically, to fast-growing and aggressive tumors. Objectives: The conditional Knock-in mouse line U2af1-Q157R will be used to study the role of the human oncogene U2AF1-Q157R in carcinogenesis. To make this mouse, the vendor needs to design and make a mouse targeting vector, microinject this vector into mouse embryonic stem cells, identify a cells clone that contain the intended targeting vector properly inserted in the U2af1 locus, use this stem cell clone to make chimera mice and test for germline transmission. This mouse model is essential to understand the mechanisms how U2AF1-Q157R expression leads to cancer. Anticipated Period of Performance: 12 months upon award Contractor's Requirements : 1. The contractor shall design and make a mouse targeting vector suitable for this project. 2. The contractor shall microinject this vector into mouse ES cells (C57BL/6 background). 3. The contractor shall identify an ES cells clone that contains the targeting vector properly inserted in the U2af1 locus. 4. The contractor shall use this stem cell clone to make chimera mice and test for germline transmission. 5. The contractor shall update the research progress every month and after each milestone is reached. Government Responsibilities: a. The government will provide DNA sequence for the U2af1-Q157R missense mutation and consult with the vendor for the project design. b. The government will not furnish any government property to the vendor. c. The government will not furnish any facilities to be used by the vendor. Reporting Requirements and Deliverables : 1. The Government will receive status update of the project every month. 2. The Government will anticipate to receive germ line transmitted F1 mice that harbor the U2af1-Q157R conditional knock-in allele after completion of the project. Inspection and Acceptance. 1. Monthly project updates via email will be accepted from the vendor. Healthy germ line transmitted F1 mice that harbor the proper U2af1-Q157R conditional knock-in allele will be accepted from the vendor. If there are any problems or questions with the mice, the vendor will be contacted via phone or email upon delivery. 2. The vendor is expected to provide monthly update about the project and the final delivery of the F1 mice that carry the intended conditional knock-in U2af1-Q157R allele. vi. Place of Performance will be held at NIH and Contractor's Location. vii. The FAR Provision 52.212-1, Instructions to Offerors -- Commercial, applies to this acquisition and is hereby incorporated by reference. All Federal Acquisition Regulations (FAR) clauses may be viewed at http://acquisition.gov/comp/far/index.html. viii. EVALUATION OF PROPOSALS: The Government will evaluate proposals that are determined technically acceptable in accordance with the Technical Evaluation factors. The Government intends to award a single contract from this solicitation. The Government reserves the right not to award a contract. Evaluation of Proposals: The Government reserves the right to make an award without discussions based solely upon initial proposals. EVALUATION FACTORS: 1. Contractor shall provide the detailed design of the targeting strategy, execute the project and ultimately provide the mouse line carrying the conditional U2AF-1-Q157R knock-in allele. A. Price (This will be Firm-Fixed Price): An evaluation of the offeror's price proposal will be made to determine if proposed prices are realistic for the work to be performed, reflect a clear understanding of the requirements, and are consistent with the technical proposal. Reasonableness determinations will be made by determining if competition exists, by comparing proposed prices with established commercial or GSA price schedules (if applicable), and/or by comparing proposed prices with the Independent Government Cost Estimate (IGCE). ix. The FAR Provision 52.212-1, Instructions to Offerors -- Commercial, applies to this acquisition and is hereby incorporated by reference. All Federal Acquisition Regulations (FAR) clauses may be viewed at http://acquisition.gov/comp/far/index.html. The proposal must reference the RFP Number: HHS-NIH-NHLBI-CSB-(HG)-2015-147-DLM. All responsible offerors may submit a proposal, which if timely received, shall be considered by the agency. All proposals shall contain the following: 1. Name of Contracting Organization; 2. The Defense Priorities and Allocations System (DPAS) and assigned rating does not apply to this acquisition. xvi.
