The hypothesis is that by understanding these interactions, and the biology of the virus, the section can contribute to the use of AAV vectors for gene therapy. The AAV Biology Section requires an Accuri C6 System W/CSampler to analyze tissues and cells for the expression of recombinant proteins. (i) "This is a combined synopsis/solicitation for commercial items prepared in accordance with the format in Subpart 12.6 as supplemented with additional information included in this notice. This announcement constitutes the only solicitation; proposals are being requested and a written solicitation will not be issued." (ii) The solicitation number is NHLBI-CSB-DE-2015-212-KEW and the solicitation is issued as a request for quote (RFQ). (iii) The solicitation document, the incorporated provisions and clauses are those in effect through Federal Acquisition Circular 2005-83 effective 03 AUG 2015. (iv) This is a small business set aside. The associated NAICS code is 334118, Computer Terminal and Other Computer Peripheral Equipment Manufacturing, and the small business size standard is 1000 Employees. The acquisition is being conducted in accordance with the procedures of FAR Part 12 and FAR Part 13. (v) The Contractor Requirements are listed below: (vi) The Contractor Shall: 1. Provide (quantity 1) Accuri C6 System W/CSampler Software and Workstation Computer - standard Optical fibers, reagent containers, C6 Flow cytometer starter kit, 1 year warranty for C6 C/Sampler, Item Number: 653119 00382906531193; specifications as follows; a. Accuri C6 Flow Cytometer System A small footprint of 11.0"H x 14.75"W x 16.5"D (27.9 x 37.4 x 41.9 cm) [11"H x 21.5"W 16.5"D with fluid bottles] and weight under 35 lbs, permits easy placement of the C6 on the lab bench, in a fume hood or a glove box. The instrument‘s compact size promotes its mobility and utilization in remote field locations. b. Must have acquisition speeds of up to 10000 events per second. The MESF for FITC <150 and PE <100. The flow rates are 14ul/ min, 35ul/min and 66ul/minute. The system also has an adjustable custom core feature which allows the user to adjust the core from 5-40 microns. This will accommodate small-large cells/particles. Resolution of smaller particles is.5 microns.Single click calibration with auto object detect c. The sheath fluid must be filtered, deionized laboratory grade water (not expensive proprietary products). The use of deionized water minimizes the cost of operation to a few cents per day, reduces corrosion due to the salt in the commercial sheath, as well as sample and reagent consumption and storage costs. Easily switch between high resolution imaging and DXA mode d. Many sample tube types can be used including 12 x 75 mm tubes, and various microcentrifuge tubes. Microcentrifuge tubes can be used for sample volumes as low as 50μL, reducing sample and reagent consumption and costs. An additional sampler can be added at any time which allows the user to automatically run tubes (20) or 96 well (deep well or regular) in a walk away mode.Images instantaneously displayed on 22" high resolution 2.3MP monitor e. Absolute cell counts can be obtained without the need to add counting beads, providing cell concentration without additional testing equipment or reagents. Sample imaging size 10cm x 15cm (4" x 6") f. A 24-bit digital signal processor generates data over a range of 7.2 decades or 16,777,215 channels and the Zoom function allows focusing on populations of interest, eliminating the need to make voltage adjustments to the detectors which, in turn, allows for easier data collection as well as no wasted samples when setting up new applications. Endogenous autofluorescence or extremely bright signals necessitate voltage adjustments on other flow cytometers in order to collect data on-scale. The wide dynamic range must allow both dim and bright signals to be displayed on the same scale. Pre-optimized and set voltage and gain settings, also enhance reproducibility of results, and comparability to data from other C6s, e,g, during collaboration.Spatial resolution 8μm at 6X geometric magnification g. No air-over-fluid pressure is required to create the hydrodynamic focusing of the sheath, therefore no aerosols are generated, providing greater safety. This also allows for very easy addition of reagents to the sample during data acquisition, without interrupting the flow. This permits superior, continuous event acquisition, for example, in calcium flux assays. Tube current 0.3mA max (isowatt limited to 12W from 41-60kV) h. A simple, quick exchange of filters is all that is required to detect additional fluorochromes. Other flow cytometers use dichroic mirrors, in addition to filters, increasing complexity and cost. Accuri offers several optional optical filters: 510 nm, 540 nm, 565 nm, 610 nm, 630 nm and 780 nm, as well as attenuation filters, that can be substituted for the standard filters supplied with the C6. Although four colors can be measured from a single experiment, with this feature, many more fluorochromes can be detected by substituting the filters.Window filtration 0.25mm Be, tungsten source i. CFlow Plus Analysis software can be used for data analysis on both a PC or on a MAC. Sample data can also be exported to third party software including FlowJo, FCS Express, WinList, etc.Dimensions external 23" W x 21" D x 36" H (59cm x 54cm x 90cm) j. The optional Selectable Laser Module can be purchased at any time and allows the choice of using three laser configurations: 1) three colors off the blue laser and one color off the red laser; 2) two colors off the blue laser and 2 colors off the red laser; and 3) four colors off the blue laser.
