- COMBINED SYNOPSIS DOCUMENTATION This is a combined synopsis/solicitation for commercial items prepared in accordance with FAR 12.6, Streamlined Procedures for Commercial Items, as supplemented with additional information included in this notice. This announcement constitutes the only solicitation; Quotes are being requested and a written solicitation will not be issued. The solicitation number is W81XWH16T0226. The solicitation is issued as a request for quotation (RFQ). Provisions and clauses in effect through Federal Acquisition Circular 2005-87 are incorporated. It is the contractor's responsibility to be familiar with the applicable clauses and provisions. The NAICS code is 325413, with a small business size of 1,000 employees. This is a Full and Open Competition requirement; all qualified vendors are encouraged to submit a quote. USAMRAA on behalf of the United States Armed Forces Medical Examiner System (AFMES) requests responses from qualified sources capable of providing equipment brand name ore equal to the Beckman Agencourt AMPure XP system 60 mL (Part Number: A63881). BRAND NAME OR EQUAL SALIENT CHARACTERISTICS: In order to optimize the new PCR amplification and sequencing techniques required for AFDIL's applications, Beckman Coulter Inc. reagents are required. These reagents will result in less re-amplifications being required, conserving both sample and cost to the government. These improvements will allow for automation, reduction of analyst error, and reduction of overall processing time. During proof of concept and development of the next generation amplification and sequencing strategies, these reagents have been proven to the government to be: reproducible from lot to lot; robust; reliable; and free from human nuclear and mitochondrial DNA contamination. The vendor's ability to utilize quality controls that produce contamination free, reproducible and reliable libraries is essential for processing forensic casework samples and meets AFDIL mission requirements. Development and optimization of the new methodologies to identify missing individuals were performed using these reagents. These new methods are currently being forensically validated in accordance with the Federal Bureau of Investigations Quality Assurance Standards, which has resulted in the need for these items. Agencourt AMPure XP System 60ml (ManfllA63881); A PCR purification system that utilizes Solid Phase Reversible Immobilization (SPRI) magnetic bead-based technology for post-PCR clean up with no salt carryover. Requires no centrifugation or filtration; can be used in manual and automated 96- well formats. Must be compatible and validated for use with specific Next Generation Sequencing library preparation kits and instruments. •Must utilize Solid Phase Reversible Immobilization (SPRI) magnetic bead-based technology for post-PCR clean up. •Must NOT require centrifugation or filtration. •Must allow for manual and automated processing in both tube and 96-well plate formats. •Must enable high recovery of PCR amplicons as well as both double and single stranded DNA templates. •Must allow for a range of fragment sizes to be recovered greater than 1OObp. •Must remove unincorporated dNTPs, primers, primer dimers, salts and other contaminants. •Must generate purified PCR products that are stable at 4°C for minimum of 7 days. •Must have reagents with storage shelf life of 12 months. •Must be compatible and validated for use with the following kits, used for Next Generation Sequencing library preparation: oNextera XT (Illumina) oNEBNext Ultra (New England Biosciences) oNEXTflex DNA Sequencing Kit (BIOO Scientific) oAccel-NGS 1S DNA Library Kit for the Illumina Platform (Swift Biosciences)
