Description
Sources Sought Automated Liquid Handling for DNA Manipulation The National Institute of Standards and Technology (NIST) seeks information on commercial vendors that are capable of providing a liquid handling laboratory instrument for DNA manipulation including next-generation sequencing library preparation, polymerase chain reaction (PCR), and DNA assembly. NIST is seeking responses from all responsible sources, including large, foreign, and small businesses. Small businesses are defined under the associated NAICS code for this effort, 334516, as those domestic sources having 1,000 employees or less. Please include your company's size classification and socio-economic status in any response to this notice. Note that 13 CFR § 125.6 is applicable to this procurement, and as such, would need to comply with the limitations on subcontracting or qualify as a non-manufacturer (see https://www.sba.gov/contracting/contracting-officials/non-manufacturer-rule/non-manufacturer-waivers). If a small business cannot meet the requirements of 13 CFR § 125.6 in response to the below requirements, it should indicate so in its response. Background and Purpose: The National Institute for Standards and Technology (NIST) needs to acquire a liquid handling laboratory instrument capable of next-generation sequencing library preparation, polymerase chain reaction (PCR), and DNA assembly. Installation and training are also required. Specifications: 1. The instrument shall be capable of running automated library preparation methods for Illumina sequencing platforms including Nextera XT DNA, 16S rRNA, TruSeq Custom Amplicon v1.5, Nextera DNA Flex, and TruSeq Stranded mRNA. 2. The instrument shall include an Eight-channel air displacement pipetting tool with the following specifications: a. For use with disposable tips b. Pipetting volume spanning at least the range from 1 uL to 1000 uL c. Independent pipetting volume for each channel d. Pipetting precision (with optimal tip size) of better than: i. 6% CV at 1 uL ii. 2% CV at 10 uL iii. 2% CV at 100 uL iv. 1% CV at 1000 uL e. Pipetting accuracy (with optimal tip size) of better than: i. 8% at 1 uL ii. 2% at 10 uL iii. 2% at 100 uL iv. 1% at 1000 uL f. Variable spacing and height (i.e. independent ‘Y' and ‘Z') for each channel g. Pressure-based liquid level detection for each channel to detect the level of liquid in SBS plates, tubes, or other labware used with the system h. Capacitance-based liquid level detection for each channel to detect the level of liquid in SBS plates, tubes, or other labware used with the system 3. The instrument shall include an automated thermocycler suitable for polymerase chain reaction in 96-well plate format with the following capabilities and specifications: a. Temperature control spanning at least the range from 4°C to 99°C b. Temperature accuracy of ±0.3°C or better at 55°C c. Temperature uniformity of ±0.2°C or better from 55°C to 95°C d. Heating rate adjustable from 0.1 to 4.4 K/s e. Cooling rate adjustable from 0.1 to 2.2 K/s f. Lid heating with temperature control spanning at least the range from 5°C above ambient to 115°C g. Compatible with 96-well skirted plates 4. The instrument shall include at least two on-deck SBS plate positions with cooling capability to 4°C or colder. 5. The instrument shall include an enclosure around the liquid handler deck area with HEPA filtration to provide positive pressure filtered air to the pipetting space within the liquid handler to protect the liquid handler deck area from the environment outside the instrument. a. The HEPA air flow shall be controllable by the instrument software. 6. The instrument shall include control software for flexible programming of new instrument methods, and shall include programmed methods for the sequencing library preparation methods listed in Specification 1. Stronger consideration shall be given to instruments for which the sequencing library preparation methods listed in Specification 1 have been qualified by Illumina to show that the resulting automatically prepared libraries perform comparably to those prepared manually. Instruction to Responders: Companies that are capable of providing this Automated Liquid Handling Instrument are requested to email a detailed report describing their abilities to Indira.Scott@nist.gov no later than the response date for this sources sought notice. The report should include achievable specifications and any other information relevant to your product or capabilities. Also, the following information is requested to be provided as part of the response to this sources sought notice: 1. Name of the company(s) that offer the required supply. In addition, provide a point of contact for the company(s). 2. Business size. 3. Indication of number of days, after receipt of order that is typical for delivery. 4. Rough estimate of pricing (if available, not required); and 5. Any other relevant information that is not listed above which the Government should consider in developing its minimum specifications and finalizing its market research.
