Purpose and Objectives The National Institutes of Health (NIH) is the nation's leading medical research agency and the primary Federal agency whose mission is to seek fundamental knowledge about the nature and behavior of living systems and the application of that knowledge to enhance health, lengthen life, and reduce illness and disability, conducting, supporting and making medical discoveries that improve people's health and save lives. The National Center for Advancing Translational Sciences (NCATS) is a translational science center that prides itself on its tremendously productive pipeline and is innovative in several ways. NCATS brings together a diverse range of scientists, including medicinal chemists, biologists, toxicologists, and engineers, in to ultimately translate basic science into real products and services that help improve people's lives. NCATS is working on a TRND Relaxin FIU project which has as goal to perform medicinal chemistry to measure type 1 collagen in a human cell line as a marker for inflammation. The Cisbio Pro-collagen type 1 (human) and Alpha-SMA assay kits are needed for this project. Project Requirements NCATS seeks to purchase twelve (12) assay kits: six (6) Pro-collagen Type 1 (human) kits and six (6) Alpha-SMA kits. NCATS seeks the below kits, or brand-name equal kits. 1. Pro-Collagen Type 1 (Human) kits Quantity: six (6) Manufacturer: Cisbio Product Code: 63ADK014PEH Salient Characteristics: • Sandwich immunoassay detecting human pro-collagen type 1 • Detectable in a sandwich assay format using two (2) different specific monoclonal antibodies, one labeled with Europium Cryptate (donor) and the second with d2 (acceptor) • Allows detection of human pro-collagen type 1 in cell supernatants • Allows quantification of human pro-collagen type 1 in cell supernatants • Allows functional screening of compounds using Human pro-collagen type 1 secreting cells • Allows for low volume sample size needed • Good for live cell, supernatant, and biochemical-based protocols • Comes as all-in-one ready to use kit with no necessity of additional purchase of components to make it work Ideally it will come with a simple "add & read" protocol, no washing step. 2. Alpha-SMA assay kits Quantity: six (6) Manufacturer: Cisbio Product Code: 62ASMAPEH Salient Characteristics: • Cell-based and able to measure expression level of alpha-SMA tissue fibrosis • Specific for alpha-SMA isoforms • Compatible with both human and mouse species • Able to identify anti-fibrotic compounds • Has high sensitivity, preferably at the endogenous level of alpha-SMA in LX-2 cell lines • Optimized for 96- and 384-well plates protocol • Good to run in both frozen cell lysates and/or fresh cell lines Anticipated Period of Performance Delivery of the kits is to occur within thirty (30) days after receipt of order. Required Information All capability statements must provide the following: (1) DUNS number; (2) organization name; (3) organization address; (4) point of contact; (5) point of contact title, address, telephone, and email address; and (6) size and type of business (e.g., 8(a), HUBZone, etc.) pursuant the applicable NAICS code.
