- The United States Department of Agriculture (USDA), Animal Plant Health Inspection Service (APHIS), has a requirement for a cell Analyzer. ������������������������������������� 2020-SS46 GENERAL INFORMATION Salient Characteristics -Reader measures microorganism growth (bacteria, molds, yeasts, or spores) automatically as defined by user -nominal voltages: 110/220 VAC 50/60 Hz -not larger than dimensions (WxDxH) 62cm x 38 cm x 23 cm -Reader functions in ambient temperature range: -25 to +50C -Reader needs temperature inside incubator range of +1 to +60C.� Must have tight temperature range for reliable results.� Temperature variations held within +/- 0.1 degree. -Incubator incorporating a liquid circulation heat exchanger to provide constant temperatures to all wells.� Essential for high quality growth curves. -Reader needs a linear shaker to assure mixing in each well. -Reader needs to measure turbidity kinetically, more sensitive than spectrophotometer. -Reader measures changes in OD vs time to produce microbiological growth curves. -Reader needs 8 filters to measure specific wavelengths: 405, 420, 450, 492, 540, 580, 600 nm and wide band.� Wide band is a �white� filter for turbidity measurement (450-580nm) -Want to run in honeycomb microplate format, 10 x 10 well plate= 100 samples/plate. -Have the ability to run two covered honeycomb plates at one time, making it possible to run 200 samples simultaneously. -No condensation or evaporation of sample. Cannot have liquid condensation on microplate lid cover.� -Microplate cover must stay on the entire experiment. -Reader needs to take turbidity measurements kinetically and automatically during the run. -Free software included -Reader needs to be able to export data to a PC and processed to generate growth curves, plotting optical density values vs time.� Ability to make 200 separate growth curves for each sample.� Data and curves need to be stored in PC until user returns. -Reader needs to be able to show users the growth curves in real time during experiment.� -Reader also needs to be able to measure up to 3 different wavelengths in one test run. -Reader software also needs to be flexible enough to pause the experiment to change experimental conditions as needed. -Reader software needs to be flexible for user to design the run conditions including: when the experiment is started, which wells to read, which filters to use, what temperature to run, amount of shaking (amplitude, speed and duration), intervals of measurements and length of run.
