(iii)�� �The solicitation document, the incorporated provisions and clauses are those in effect through Federal Acquisition Circular 2020-05 effective 27 FEB 2020. (iv)�� � The associated NAICS code is 334516, Analytical Laboratory Instrument Manufacturing, and the small business size standard is 1,000. �The acquisition is being conducted in accordance with the procedures of FAR Part 12 and FAR Part 13.� (v)�� �The Contractor Requirements are listed below: 1.0�� �PURCHASE ORDER TITLE SpectraMax iD3 Multi-Mode Microplate Reader with injectors �� ��� � 2.0�� �BACKGROUND Our ongoing research at the NIDDK studies biological functions of thyroid hormone receptor beta gene (Thrb) that requires to make next generation sequencing libraries using tiny amount of RNA from either single cell or small tissue including retina/cochlear sensory neurons, adrenal gland and pituitary. Our current research topics include the study of specific selection of these small-population cells or developmental transcriptome analyses from Thrb-expressing tissues/cells. Single-cell sequencing technique, which is powered by recently developed next-generation sequencing (NGS), enables the study of transcription factor binding site, transcriptomic analysis, epigenomic modification changes and chromatin structure. Our lab developed a single cell capturing technique from tiny TRb-population tissue to investigate transcriptomic analysis of individual Thrb positive cells. Among series procedures involved in the RNA-seq library construction, the step of cDNA fragmentation is critical, in which evenly and efficiently sonication of the cDNA strands into desired size is the key factor to ensure high quality libraries and optimum data. We are trying to identify Thrb true target genes using transcriptional activity assay, Western blot, ELSA assay and so on. �� 3.0�� �OBJECTIVE We would like to purchase the SpectraMax iD3 Multi-Mode Microplate Reader with injectors to measure transcriptional activity by luciferase reporter assay, RNA, DNA and protein concentration from small tissue for investigating true target genes of thyroid hormone receptor beta. This SpectraMax iD3 Multi-Mode Microplate Reader with injectors provides various wavelength range including absorbance wavelength range (230~1000nm), fluorescence wavelength range (exited 250~830nm, emitted 270~850nm) and luminescence wavelength range (300~850nm) and multi-mode read types including endpoint, kinetic, spectrum scan and well scan. This device uses a monochromater which allows for extended wavelength capability without a lot of individual filters. iD3 has ultra-coold photomultiplier tube (PMT) which is reduced background caused by heat, allowing the instrument to produce more accurate data. This equipment fits our requirements on reduction of processing times, sample input flexibility, accurate multi-mode detection.� 4.0 �� �SALIENT / REQUIRED FEATURES AND SPECIFICATIONS 1.�� �This instrument must have multi-functions including absorbance, fluorescence, and luminescence read modes. It also must possess integrated linear, orbital, and double orbital shaking capabilities and temperature control up to 66 degrees. This instrument must be suitable for various needs of researchers and is convenient for performing various experiments to get a precious data.� 2.�� �A monochromator with a wavelength range of 250nm-1000nm will be used in place of filters enabling the user to adjust wavelength by 1nm increments and eliminating the need for filters. In addition, the instrument must have a patented ultra-cooled photomultiplier tube (PMT) which reduces background caused by heat, allowing the instrument to produce more accurate data. This is important to measure the accurate values of our various assay for downstream process. 3.�� �This instrument must be configured with SmartInjectTM dual injector module that allows for simultaneous injection and mixing to capture rapid signal development. The injector module also includes an overflow detector and produces a significantly low dead volume minimizing loss of reagent. 5.0 �� �TASKS AREAS 5.1�� �Task Area 1 �The Contractor will deliver the new SpectraMax iD3 Multi-Mode Microplate Reader with injectors on site to Building 10. Delivery is to be inside delivery, to the area where the SpectraMax iD3 Multi-Mode Microplate Reader with injectors is to be installed. � 5.2�� �Task Area 2 � Vendor Technician shall install the new SpectraMax iD3 Multi-Mode Microplate Reader with injectors in the lab. � 5.3 � � � Task Area 3 � Vendor will test the new SpectraMax iD3 Multi-Mode Microplate Reader with injectors and provide training for the lab within thirty days of installation. Vendor will help to set up new protocols to optimize conditions for our lab�s samples.� 6.0�� �PURCHASE ORDER TYPE This Purchase Order is to be a firm fixed price.� 7.0�� �INSPECTION AND ACCEPTANCE Inspection and acceptance will be performed at the place of installation - National Institutes of Health, NIDDK, 9000 Rockville Pike, Maryland 20892. (vi)�� �This requirement is being pursued through the authority granted in FAR 6.302-1(c) Application for Brand Name Descriptions. This is a requirement for a Molecular Devices SpectraMax iD3 Multi-Mode Microplate Reader with injectors to enable the NIDDK to measure transcriptional activity by luciferase reporter assay, RNA, DNA and protein concentration from small tissue for investigating true target genes of thyroid hormone receptor beta. (vii)�� �Period of Performance: �Within 60 Days After Date of Award (viii)�� �The provision at FAR clause 52.212-1, Instructions to Offerors � Commercial Items, applies to this acquisition.
