Description
���� Laboratory mice are common experimental animals in biological, medical, pharmacological and psychological researches primarily because they are easy to maintain and reproduce quickly Dissection of brain tissue is an important step in sample preparation for (subcellular) proteomics studies. Some Labs (vendors) can perform brain removal and separate dissection of multiple brain regions from a single brain and follow a step-by-step protocol. This concerns dissection from fresh or frozen tissue of cerebellum, hippocampus, prefrontal cortex, and striatum for protein immunoblot and/or neuronal culture, providing technical reference information for minimizing the number of animals used in projects, and refining methods and procedures to quick brain dissection, embedding and staining.� NeuroScience Associates is one of the rare companies that can perform all stages in one Lab over a period of time with consistent and accurate results. � The Lab needs to investigate the effect of overexpression of carboxypeptidase E-neurotrophic factor 1 in the hippocampus on prevention or treatment of neurodegeneration and cognitive dysfunction in the Alzheimer's Disease mouse model�, and proposes to inject AAV-CPE into mouse brain at 2 month of age and evaluate the pathophysiological and behavioral changes at 6-7 months of age, to study whether CPE has prevented the development of AD. Also, they will inject AAV-CPE into mouse brain at 4 months of age when there is intracellular accumulation of A?, and pathophysiological and behavioral changes evaluated at 6 -7 months of age, to determine whether CPE has any therapeutic effect. the end of our treatment of mice, the Lab will provide 50 mouse brains to the company from the experiments. By using MultiBrain� technology, 25 mouse brains will be embedded together into a single block, freeze-sectioned at 25? in the coronal plane through the whole mouse brain. All sections that are cut will be collected into a series of 24 cups containing antigen preserve solution. Thionine Nissl stain and several antibody stains will be performed through the cortex/cerebrum (hippocampus and pre-frontal cortex) on every 12th section spaced at 300? intervals, yielding ~30 2 x 3 slides (~30 stained sections per mouse brain, ~750 stained sections total) per stain. Approximately 30 days after the shipment of the stained slides, unstained sections for each block will be sent to NIH in antigen preserve solution, the total number of unstained sections to be sent is ~13500. ��These services have been provided in the past by one main vendor who can do dissecting, embedding and staining all in one Lab without the need to sub-contract one of the services� This proposed procurement is for a service for which the government intends to solicit and negotiate on a sole source basis to NeuroScience Associates located in Knoxville, TN. 37934. �In order to comply with the requirements in this Special Notice, the responses should be based on our interpretation of the general requirements provided below. Note 22. The proposed contract action is for a continuation of previous services for which the Government intends to solicit and negotiate with only one source under the authority of FAR 6.302. Interested persons may identify their interest and capability to respond to the requirement or submit quotations. This notice of intent is not a request for competitive quotes. However, all quotes received within the time and date of publication of this ""Special Note 22"" will be considered by the Government. A determination by the Government not to compete with this proposed contract based upon responses to this notice is solely within the discretion of the Government. Information received will normally be considered solely for the purpose of determining whether to conduct a competitive procurement. HOW TO RESPOND: In order to compete for this project, interested parties must be registered in the Central Contracting Registration and submit a price quotation by 11:00 am, ET, July 17th 2020.
