- DESCRIPTION: The National Institutes of Health - NICHD intends to procure the services of a contractor to provide synthesis and Cloning of 101 & 218 Individual Genes to determine the binding of various mouse Znf alleles alone and to each other and when co-expressed with mPRDM9.� There are 319 different but highly repetitive codon optimized mouse znf arrays cloned into an expression vector. �Lab�s rationale for sole source: ��We study how the epigenetic state of mammals is initially established in early development, by exploring KRAB-zfp transcription factors that recognize parasitic mobile DNA elements to initiate stable heterochromatic silencing. We also study how one large family of these parasitic elements, the endogenous retroviruses or ERVs, play an important role in development and evolution of new traits in mammals by rewiring gene expression networks.� Finally, we explore how the epigenetic state of a cell works in conjunction with combinatorial codes of transcription factors to generate the huge diversity of cell types required for mammalian development. These constructs will be used to elucidate how these transcription factors interact with chromatin and regulate the expression of genes, by expressing in cells and then doing ChIP-seq to look at binding properties. This company has previously synthesized all the ZFP arrays used in our labs prior and current ongoing experiments. Changing variables in the middle of the project/experiments would compromise the integrity of the previous data collected.� These newly synthesized arrays would be used in conjunction with the ones this company has previously produced to maintain continuity of the projects in the lab. All the SC1692 VectorArk Cloning is free of charge instead of $49.00 each, saving thousands for the Lab which we will not receive from any other vendor, assuming that all cloning being equal This proposed procurement is for a service for which the government intends to negotiate with one source, Genscript USA, Inc located in Piscataway, NJ.�� In order to comply with the requirements in this Special Notice, the responses should be based on our interpretation of the general requirements provided.� Interested persons may identify their interest and capability to respond to the requirement or submit quotations. This notice is not a request for competitive quotes. However, all quotes received within the time and date of publication of this ""Special Notice� will be considered by the Government. A determination by the Government not to compete with this proposed contract based upon responses to this notice is solely within the discretion of the Government. Information received will normally be considered solely for the purpose of determining whether to conduct a future competitive procurement.
