- Modified to add the following:��NOTE TO CONTRACTOR: The contractor shall provide a completed copy of FAR 52.204-24, ""Representation Regarding Certain Telecommunications and Video Surveillance Services or Equipment"" with their quote in response to this solicitation.� SEE ATTACHMENT.� �There is no other change to this notice. This is a combined synopsis/solicitation for commercial items prepared in accordance with the format in FAR 12.6 as supplemented with additional information included in this notice.� This announcement constitutes the only solicitation and a separate written solicitation will not be issued.� This solicitation number is NICHD-20-240 and is issued as a Request for Quotation (RFQ).� The solicitation/contract will include all applicable provisions and clauses in effect through Federal Acquisition Circular 2020-08.� The North American Industry Classification (NAICS) Code is 541380 and the business size standard is $16.5m.� However, this solicitation is not set aside for small business.� This acquisition is being conducted using Simplified Acquisition Procedures in accordance with FAR Part 13.� Eunice Kennedy Shriver National Institute of Child Health and Human Development (NICHD) intend to procure on a sole source basis with Cyagen US of Santa Clara, CA to provide service for generation of conditional mouse models. �Services/work to be provided broken down into four projects.� See Attached.�� For conditional knockin/out, the lab @ NIH will be able to breed founders directly to their tissue-specific Cre mice, which allows them to advance their research much more quickly than conventional knock-out generation. Generation of a conditional mouse line requirements are: Supports vector design, construction, validation, and sequencing. Targeting vector will be injected into ES cell line that generates 100% ESC-derived founder mice rather than chimeras, eliminating the need to screen for germline transmission.� Clones are then screened by drug selection and isolation of drug resistant clones. Positive ES clones are then further validated by PCR, Southern blotting, and chromosome counted (for correct number of chromosomes). ES clones are then injected into mouse host embryos and transferred into surrogate mothers. A selection cassette on the targeting construct capable of removing itself after ES cell targeting without the need to breed to Flp deleter mice, thus bypassing F1 mouse generation.� These innovations eliminate two generations of breeding, shortening production time by 4-6 months as compared to industry standard. � Cyagen Biosciences is, to the best of our knowledge, the sole source for: (a) the accelerated ES-cell based methodology for conditional mouse model generation known as TurboKnockout. ��Cyagen proprietary TurboKnockout� service can provide a conditional knockout, reporter knockin and humanization mouse model as fast as 6 months. This proprietary gene targeting method is enabled by two innovations: (1) A super competent ES cell line that generates 100% ESC-derived founder mice rather than chimeras, eliminating the need to screen for germline transmission. (2) A selection cassette on the targeting construct capable of removing itself after ES cell targeting without the need to breed to Flp deleter mice. These innovations eliminate two generations of breeding, shortening production time by 5-6 months as compared to industry standard. (b) the associated guarantee covering TurboKnockout� based projects ensuring that a minimum of three (3) germline transmitting heterozygous mice will be delivered upon project completion.� Cyagen� s animals are maintained in AAALAC-accredited SPF barrier facilities. Animal health is monitored by a stringent sentinel program. Sentinel animals are tested for a list of excluded pathogens. Cyagen guarantee that their animals are free of all the pathogens on the exclusion list. All animal work performed at Cyagen complies with all federal/national and state/provincial regulations governing animal care and use in the US and China.� Cyagen strive to maintain an animal welfare standard that meets and frequently exceeds guidelines set forth by AAALAC and the Office of Laboratory Animal Welfare (OLAW) of the NIH. � The proposed contract action is for supplies or services for which the Government intends to solicit and negotiate with only one source under authority of the 41 U.S.C. 253(c) (1), FAR 6.302.� Only one responsible source and no other supplies or services will satisfy agency requirements.
