- This is a Small Business Sources Sought notice. This is NOT a solicitation for proposals, proposal abstracts, or quotations. The purpose of this notice is to obtain information regarding: (1) the availability and capability of qualified small business sources; (2) whether they are small businesses; HUBZone small businesses; service-disabled, veteran-owned small businesses; 8(a) small businesses; veteran-owned small businesses; woman-owned small businesses; or small disadvantaged businesses; and (3) their size classification relative to the North American Industry Classification System (NAICS) code for the proposed acquisition. (4) availability of domestic sources manufactured in the United States in sufficient and reasonably available commercial quantities and of a satisfactory quality. For equipment/supply requirements, small business responses must include: (1) the place of manufacturing (i.e. address if supply/equipment is a domestic end product and include country of manufacture), as well as (2) the size status of the manufacturer under the applicable NAICS code (i.e. address Non-Manufacturer Rule). Your responses to the information requested will assist the Government in determining the appropriate acquisition method, including whether a set-aside is possible. An organization that is not considered a small business under the applicable NAICS code should not submit a response to this notice. Background and Purpose and Objectives: The National Institute on Aging (NIA), Translational Geroproteomics Unit of The Translational Gerontology Branch requires the use of an Orbitrap Eclipse Mass Spectrometer will be used to keep up with demands for proteomic research across TGB, LGG, LMBI, LEPS and LCI laboratories within the NIA IRP. The Eclipse Mass Spectrometer is a high versatile workhorse mass spectrometer uniquely capable of performing high throughput �discovery� analysis of protein samples with unparalleled proteome coverage and quantitative power in an unbiased untargeted manner. This instrument is capable of highly reproducible and robust �data-independent acquisition� workflows employed by a growing number of projects at the NIA. The Orbitrap Eclipse is also capable of highly multiplexed and sensitive targeted �SureQuant� assays, which take internal standard triggered peptides to perform the most sensitive and quantitative analysis of multiple peptides in highly challenging sample matrices such as plasma that are utilized across multiple studies in the NIA such as SLAM and BLSA. Salient Characteristics: The Mass Spectrometer system with nano-electrospray ionization source and on-line flow (all-in-one nano-, capillary-, and micro-) High Performance Liquid Chromatography system and automated sample injector. This integrated platform combines quadrupole, linear ion trap, and Orbitrap mass analyzers and features CID, HCD, ETD and EThcD fragmentation techniques, with subsequent mass analysis in either the ion trap or Orbitrap mass analyzer. The new generation Orbitrap Fusion Eclipse with ETD and FAIMS Pro is an extremely efficient platform for proteomics applications to perform fast MS/MS sampling at very high resolution, and it has exceptional sensitivity and superior dynamic range. The Lumos technical features and possible workflows are unique among high end mass spectrometry platforms, in that they combine novel FAIMS Pro ion mobility gas phase separation technologies and the various Quantitative Proteomics Approaches possible using isobaric labeling and multiplexing FAIMS-TMT and TMT-SPS-MS3, as well as label-free approaches FAIMS-LFQ (MS1-based label free quantification from data-dependent acquisitions, DDA) as well as label free data-independent acquisitions (DIA). Key capabilities: � Mass Spectrometer to combine the state of the art of quadrupole, Orbitrap, and linear ion trap mass analysis in a revolutionary architecture. It is the only instrument available that has combined, simultaneous, 450,000 resolving power, with attomole sensitivity. � Double to acquisition rates (40hz +, for the orbitrap and linear ion traps) of current instrumentation in the currently available Q-Exactive High Field mass spectrometer (QE-HF) � Orbitrap resolution up to 1 million at 200m/z versus 240,000 on the current QE-HF instrument � Uniquely capable of advanced PTM studies with multiple scan types including ETD, HCD and �combined� EThcD fragmentation for increased peptide sequence coverage and improved PTM-site localization. Additional fragmentation options also improve identifications of labile post-translational modifications such as phosphorylation, molonylation, and glycosylation. � Highly multiplexed isobaric labeling with tandem mass tagging (TMT, up to TMTpro-18plex) to greatly reduce instrument time for sample analysis. � Ion mobility capabilities with FAIMS FAIMS improves proteome coverage significantly in complex tissue lysates and, in particular, high dynamic range biofluids such as plasms. � Gas phase fractionation with FAIMS can greatly enhance the depth of coverage with data-independent acquisition methods currently employed by the proteomics laboratory. � Unique capable of highly sensitive targeted assays based on the SureQuant internal standard (IS) targeted protein quantitation workflow. SureQuant is a novel targeted PRM workflow that leverages internal standards to guide and automatically maximize outcomes and data quality for real-time targeted proteomics analysis and maximize the detection of very low abundance peptides in high dynamic range samples such as plasma. � Extends the mass range to m/z 8000 for detection of precursor and product ions in the Orbitrap mass analyzer � More efficient precursor selection, as little as 0.4m/z precursor isolation widths � Flexible, expandable platform for the qualitative and quantitative analysis of proteomics data with a modular design of customized analysis pipelines that incorporate a variety of commonly used proteomic analysis workflows and algorithms. Anticipated Delivery Date: 14-15 weeks after Award of Order (ARO). Capability statement /information sought: Small business concerns that believe they possess the capabilities to provide the required products should submit documentation of their ability to meet each of the project requirements to the Contracting Officer. The capability statement must specifically address each of the project requirements separately.� Additionally, the capability statement should include 1) the total number of employees, 2) the professional qualifications of personnel as it relates to the requirements outlined, 3) any contractor GSA Schedule contracts by which all of the requirements may be met, if applicable, and 4) any other information considered relevant to this program. Small businesses must also provide their Company Name, DUNS number, Physical Address, and Point of Contact Information. Interested small businesses are required to identify their type of business, applicable North American Industry Classification System Code, and size standards in accordance with the Small Business Administration. The government requests that no proprietary or confidential business data be submitted in a response to this notice. However, responses that indicate the information therein is proprietary will be properly safeguarded for Government use only. Capability statements must include the name and telephone number of a point of contact having authority and knowledge to discuss responses with Government representatives. Capability statements in response to this market survey that do not provide sufficient information for evaluation will be considered non-responsive. When submitting this information, reference the solicitation notice number. One (1) copy of the response is required and must be in Microsoft Word or Adobe PDF format using 11-point or 12-point font, 8-1/2� x 11� paper size, with 1� top, bottom, left and right margins, and with single or double spacing. The information submitted must be in an outline format that addresses each of the elements of the project requirements and in the capability statement /information sought paragraphs stated herein.� A cover page and an executive summary may be included but is not required. The response is limited to ten (10) pages.� The 10-page limit does not include the cover page, executive summary, or references, if requested.
