- Notice of Intent to Sole Source for 10X Genomics NIH-SS-6320313 The National Institutes of Health intends to award a Firm-Fixed-Price purchase order to 10x Genomics, Inc., 6230 Stoneridge Mall Road, Pleasanton, California, 94588.� �� In accordance with FAR Part 6.302-1, the Contractor is the only known responsible source who can provide the subscription service as no other source has been identified who can provide the requirement listed below. We are ordering the following 10X Genomics kits: Chromium Next GEM Chip J Single Cell Kit Chromium Next GEM Single Cell Multiome ATAC + Gene Expression Reagent Bundle Chromium Next GEM Single Cell 3' GEM, Library & Gel Bead Kit v3.1 Chromium Next GEM Chip G Single Cell Kit Chromium Single Cell 3� Feature Barcode Library Kit The Chromium Single Cell Gene Expression Solution upgrades short read sequencers to deliver a scalable microfluidic platform for 3? digital gene expression by profiling 500-10,000 individual cells per sample. A pool of ~3,500,000 10x Barcodes are sampled separately to index each cell�s transcriptome. It is done by partitioning thousands of cells into nanoliter-scale Gel Beads-in-emulsion (GEMs), where all generated cDNA share a common 10x Barcode. Libraries are generated and sequenced from the cDNA and 10x Barcodes are used to associate individual reads back to the individual partitions. The Chromium Next GEM Single Cell 3� Reagent Kits v3.1 provide: High gene sensitivity, low multiplet rates Compatible with Feature Barcoding technology for Cell Surface Protein profiling or CRISPR Screening Robust workflow Simple chip loading Compatible with whole cells and nuclei Industry leading cell capture rates (~65%) Broad range of sample throughput (1-8 samples per chip) Broad range of cell throughput per sample (500 - 10,000 cells) Cell size flexibility - no lower limits Chromium Single Cell v3.1 uses: Chromium Next GEM Single Cell 3� GEM, Library & Gel Bead Kit v3.1, 16 rxns, 1000121 Chromium Next GEM Chip G Single Cell Kit, 48rxns, 1000120 Chromium Single Cell 3� Feature Barcode Library Kit, 16 rxns, 1000079 Chromium Single Cell Multiome + Gene Expression provides a comprehensive scalable multiomic approach for simultaneously profiling epigenomic landscape and gene expression in the same single nuclei. This is achieved by transposing nuclei in a bulk solution. Using a microfluidic chip, the nuclei are partitioned into nanoliterscale Gel Beads-in-emulsion (GEMs). A pool of 736,000 10x Barcodes is sampled to separately and uniquely index the transposed DNA and cDNA of each individual nucleus. ATAC and gene expression (GEX) libraries are generated from the same pool of pre-amplified transposed DNA/cDNA and sequenced. The 10x Barcodes in each library type are used to associate individual reads back to the individual partitions, and thereby, to each individual nucleus. Chromium Next GEM Single Cell Multiome ATAC + Gene Expression uses: Chromium Next GEM Single Cell Multiome ATAC + Gene Expression Reagent Bundle, 16 rxns, 1000283 Chromium Next GEM Chip J Single Cell Kit, 48 rxns, 1000234 All above solutions and supplies are provided by 10X Genomics specifically to fit the kits. This notice is not a request for competitive quotes.� However, any party that believes it can provide the products as stated herein may submit a written capability statement that clearly supports and demonstrates their ability to perform the requirement. Responses to this notice shall contain sufficient information to establish the interested parties� bona-fide capabilities for fulfilling the requirement and include: unit price, list price, shipping and handling costs, the delivery period after contract award, the prompt payment discount terms, the F.O.B. Point (Destination or Origin), the Dun & Bradstreet Number (DUNS), the Taxpayer Identification Number (TIN), and the certification of business size.� A determination by the Government to compete this proposed award based upon responses to this notice is solely within the discretion of the Government. In order to be considered for an award, an offeror must have completed the online electronic Representations and Certifications located at https://www.sam.gov in accordance with FAR 4.1201(a). By submission of an offer, the offeror acknowledges the requirement that a prospective awardee shall be registered in SAM at https://www.sam.gov/ prior to award, during performance, and through final payment of any contract, basic agreement, basic ordering agreement, or blanket purchasing agreement resulting from this solicitation (Lack of registration in the System for Award Management will make an offeror ineligible for award).� All responsible sources may submit an offer that may be considered by the Agency. It is anticipated that an award will be issued approximately five (5) days after the date of this notice unless the Government determines that another organization has the capability to meet this requirement.
