Description
(i)� This is a combined synopsis/solicitation for commercial items prepared in accordance with the format in Subpart 12.6 as supplemented with additional information included in this notice. This announcement constitutes the only solicitation; proposals are being requested and a written solicitation will not be issued. (ii)� The solicitation number is 75N95022R00093 and the solicitation is issued as a request for quotation (RFQ). This acquisition is for a commercial item or service and is conducted under the authority of the Federal Acquisition Regulation (FAR) Part 13�Simplified Acquisition Procedures; �and FAR Part 12�Acquisition of Commercial Items, and is not expected to exceed the simplified acquisition threshold. THIS IS A NON-COMPETITIVE (NOTICE OF INTENT) COMBINED SYNOPSIS SOLICITATION TO AWARD A CONTRACT OR PURCHASE ORDER WITHOUT PROVIDING FOR FULL OR OPEN COMPETITION (INCLUDING BRAND-NAME).� The National Institute on Drug Abuse (NIDA), Office of Acquisition (OA), on behalf of the National Center for Advancing Translational Sciences (NCATS), intends to negotiate and award a Firm Fixed Priced contract without providing for full and open competition (including brand-name) to University of North carolina at Chapel Hill.,104 Airport Dr. Ste 2200, Chapel Hill NC, 27599 for purified recombinant (GHSR) for use in cryoEM structural determination studies. This acquisition is conducted as non-competitive for a commercial item or service and is conducted pursuant to FAR 13.106-1(b)(1). This acquisition is restricted to UNC-CH Protein Expression & Purification and Macromolecular Crystallography Core Lab (PEP-MX). The rationale for the sole source justification is that The University of North Carolina (UNC) has already developed and has an established protocol to run the protein production. The project consists of the cryo-electron microscopy (cryo EM) structure determination of ghrelin receptor (hGHSR) preferably in a large stable complex with native binding partners and stabilizing proteins. There are no other vendors in the market now that have a well-established protocol for this type of protein. Sourcing this project to a different vendor at this stage of the project will mean doing double work as previously collect data would be lost and unusable for comparisons. Additionally changing vendors would lead to several months of delay in cryoEM experiments that are critical for advancing the program. (iii)� The solicitation document and incorporated provisions and clauses are those in effect through Federal Acquisition Circular (FAC) Number 2022-06 dated May 26, 2022. (iv)� The associated NAICS code 541990, All Other Professional, Scientific and Technical Services and the size standard in millions of dollars is $16.5. (v)� This requirement is to acquire purified recombinant (GHSR) for use in cryoEM structural determination studies. Background: The National Center for Advancing Translational Sciences (NCATS), the NCATS Early Translation Branch (ETB) collaborates with investigators at NIH and in the academic, biopharmaceutical and nonprofit sectors to generate probes for studying a diverse cross-section of human biology, focusing specifically on new targets and untreatable diseases. The Growth hormone secretagogue receptor (GHSR), also known as ghrelin receptor, is a G-protein coupled receptor that is expressed in different tissues, with high expression in brain subregions.� GHSR binds to growth hormones (e.g. ghrelin) to regulate food intake and body weight.� Pharmacological modulation of the receptor has also been implicated in aging and addiction. NCATS has developed small molecule biased agonists of GHSR that alter dopamine release in the brain and are of interest for brain disorders involving dopamine dysfunction. This requisition is to produce recombinant GHSR protein for use in cryoEM, which will allow the project team to precisely define the binding of the small molecule to the receptor. Purpose: This requisition is to acquire purified recombinant (GHSR) for use in cryoEM structural determination studies.� Project Requirements: Independently and not as an agent of the Government, the Contractor shall furnish all the necessary services, qualified personnel, material, equipment, and facilities, not otherwise provided by the Government as needed to perform the below: Nb35 production.� Nb35 is a nanobody that stabilizes GPCR:Gs complexes.� The vendor will produce Nb35 in bacteria using an expression plasmid bearing a gene for Nb35 targeted to the periplasm (4L culture).� Nb35 will be purified following bacterial lysis, NiNTA affinity capture, ion exchange chromatography, and dialysis. Expression of HA-His-Bril-hGHSR-LgBiT, Gq, Gb1-HiBiT, Gg2, and Ric8a.� The vendor will co-express the five expression vectors in HEK293 (Expi293 expression system).� Cells will be lysed in aqueous buffer and Nb35 will be added. ScFv16 may be required in addition to or in place of Nb35. Apryase, ghrelin, or N8279 may also be required.� The requirements will be selected based on experimental data and in consultation with ETB and its collaborators.� NiNTA purification will be performed in the presence of LMNG and CHS.� Size exclusion chromatography will be performed using Sephacryl S-200 with buffer containing LMNG, GDN, and CHS.� Protein quantity and quality will be determined using SDS-PAGE. Scientists with specialized training membrane protein expression and purification conduct the experiment under the supervision of the main investigator. Deliverables Contractor shall deliver a purified sample of the GHSR complex, 100 ul or more at a concentration of 5 mg/ml or higher. Reporting Requirements The vendor will assess protein purification at various stages of the process via SDS PAGE.� A final QC SDS PAGE validation will be performed and sample delivered to ETB or ETB-desired recipient. (vi)���The Government anticipates award of a firm fixed-price contract for this acquisition, and the anticipated period of performance to be six (6) months after receipt of order (ARO) (vii)� The provision at FAR 52.252-1, Solicitation Provisions Incorporated by Reference (Feb 1998), applies to this acquisition.
