B -- PRODUCTION OF SEED VIRUS POOLS SOL N02-9905 DUE 030499 POC Sherna Brown, PA, 301-402-2282/Carolyn Sentz, Contracting Officer 301-402-2282 This is a notice of intent to procure a production of seed virus pools from Dyncorp. The purpose of this contract is to propagate, or continue growing, the human x bovine rotavirus reassortments in FRHL-2 cell cultures with the specific aim to obtain yields on the order of 10-7 pfu/ml. These seed virus pools must have a titer of approximately 10-7 PFU/Ml in FRHL-2 (Fetal Rhesus Diploid Lung) cells for each of the following 6 human x bovine rotavirus candidate vaccines: DxUK, DS-1xUK, PxUK, ST3xUK, AU32xUK, WaxUK. These pools will be tested for sterility and infectivity as needed. Pools will be tested at the NIH as needed and the final results for titer will be confirmed. Dyncorp has produced all of the seed virus pools for the experiments requiring this vaccine, which will attempt to continue growing this viruses at a higher volume. Because Dyncorp has provided the vaccines required in the studies, it is imperative for the sake of consistency and so not to introduce any unknown variables that Dyncorp also manufacture further vaccine material.
